The Sanger method is based on sequentially synthesising a DNA strand complementary to a single strand (used as a template) in the presence of DNA polymerase, the four 2′-deoxynucleotides that form the DNA sequence (dATP, dGTP, dCTP and dTTP) and four dideoxynucleotides.
The use of primers specific for known genes can detect different polymorphisms in the sequence.
